RESUMO

Testing the feasibility of soil phytoremediation requires the development of models applicable on a large scale. Phytoremediation mechanisms include advanced rhizosphere biodegradation, phytoaccumulation, phytodegradation, and phytostabilization. The aim of this study was to evaluate the phytoremediation potential of the Sinapis alba. Identification of the factors influencing the extraction process of metals from contaminated soils in a laboratory system suitable for evaluating the phytoavailability of these metals in three solutions (M1-CaCl2, M2-DTPA, and M3-EDTA) included the following: distribution of metals in solution (Kd), soil properties and mobile fractions (SOC, CEC, pH), response surface methodology (RSM), and principal component analysis (PCA). The evaluation of the phytoremediation potential of the Sinapis alba plant was assessed using bioaccumulation coefficients (BACs). The accumulation of heavy metals in plants corresponds to the concentrations and soluble fractions of metals in the soil. Understanding the extractable metal fractions and the availability of metals in the soil is important for soil management. Extractable soluble fractions may be more advantageous in total metal content as a predictor of bioconcentrations of metals in plants. In this study, the amount of metal available in the most suitable extractors was used to predict the absorption of metals in the Sinapis alba plant. Multiple regression prediction models have been developed for estimating the amounts of As and Cd in plant organs. The performance of the predictive models generated based on the experimental data was evaluated by the adjusted coefficient of determination (aR2), model efficiency (RMSE), Durbin-Watson (DW) test, and Shapiro-Wilk (SW) test. The accumulation of the analyzed metals followed the pattern Root > Pods > Leaves > Seeds, stems > Flowers for As and Leaves > Root > Stem > Pods > Seeds > Flowers for Cd in soil contaminated with different metal concentrations. The obtained results showed a phytoremediation potential of the Sinapis alba plant.

In the last decades, increased intakes of contaminants and the habitats' destruction have produced drastic changes in the aquatic ecosystems. The environmental contaminants can accumulate in aquatic organisms, leading to the disturbance of the antioxidant/prooxidant balance in fish. In this context, we evaluated the level of organic, inorganic and microbiological pollutants in four leisure lakes (Chitila, Floreasca, Tei and Vacaresti) from Bucharest, the largest city of Romania, in order to compare their effects on hepatopancreas and gills metabolism and antioxidant defense mechanisms in Carassius gibelio, the most known and widespread freshwater fish in this country. The lowest level of oxidative stress was recorded in the case of fish collected from the Vacaresti lake, a protected wetland area where aquatic organisms live in wild environmental conditions. In contrast, significant oxidative changes were observed in the hepatopancreas and gills of fish from the Chitila, Floreasca and Tei lakes, such as reduced glutathione S-transferase activity and glutathione level, and increased degree of lipid peroxidation, being correlated with elevated levels of pesticides (such as 2,4'-methoxychlor) and Escherichia coli load in these organs. Although different patterns of pollutants' accumulation were observed, no important interindividual variations in cytosine methylation degree were determined. In conclusion, the presence and concentrations of metals, pesticides and antibiotics varied with the analyzed tissue and sampling site, and were correlated with changes in the cellular redox homeostasis, but without significantly affecting the epigenetic mechanisms.

The paper presents the validation of an electrochemical procedure for on-site Hg2+ ions determination in wastewater samples using a modified carbon screen-printed electrode (SPE) with a complexing polymeric film based on poly(2,2'-(ethane-1,2-diylbis((2-(azulen-2-ylamino)-2-oxoethyl)azanediyl))diacetic acid) (polyL). Using metal ions accumulation in an open circuit followed by anodic stripping voltammetry, the SPE-polyL electrode presents a linear range in the range of 20 µg/L to 150 µg/L, with a limit of detection (LOD) = 6 µg/L, limit of quantification (LOQ) = 20 µg/L, and an average measurement uncertainty of 26% of mercury ions. The results obtained in situ and in the laboratory using the SPE-polyL modified electrode were compared with those obtained by the atomic absorption spectrometry coupled with the cold vapor generation standardized method, with the average values indicating excellent recovery yields.

Fish are able to accumulate by ingestion various contaminants of aquatic environment, with negative consequences on their intestine, being continuously threatened worldwide by heavy metals, pesticides and antibiotics resulted from the human activities. Consequently, the health of other species can be affected by eating the contaminated fish meat. In this context, our study aimed to perform a comparison between the changes in intestine samples of Carassius gibelio individuals collected from different artificial lakes in Bucharest (Romania), used by people for leisure and fishing. The presence of various metals, pesticides and antibiotics in the gut of fish was assessed in order to correlate their accumulation with changes of antioxidative enzymes activities and microbiome. Our results showed that fish from Bucharest lakes designed for leisure (Chitila, Floreasca and Tei lakes) have an increased level of oxidative stress in intestine tissue, revealed by affected antioxidant enzymes activities and GSH levels, as well as the high degree of lipid peroxidation, compared to the fish from protected environment (Vacaresti Lake). Some heavy metals (Fe, Ni and Pb) and pesticides (aldrin and dieldrin) were in high amount in the gut of fish with modified antioxidative status. In conclusion, our study could improve the knowledge regarding the current state of urban aquatic pollution in order to impose several environmental health measures.

This study presents the behavior of white mustard seedlings Sinapis alba grown for three months in laboratory polluted soil containing As, Cd, Ni and Pb. Four different experiments were performed in which As was combined with the other three toxic metals in different combinations (As, AsCd, AsCdNi, AsCdNiPb), keeping the same concentrations of As and Cd in all tests and following the national soil quality regulations. The effects of these metals were monitored by the analytical control of metal concentrations in soil and plants, bioavailability tests of mobile metal fractions using three different extracting solutions (DTPA + TEA + CaCl2-DTPA, DTPA + CaCl2-CAT, and CH3COONH4 + EDTA-EDTA) and calculation of bioaccumulation and translocation factors. Additionally, micro, and macro-nutrients both in soil and plant (root, stem, leaves, flowers and seeds) were analyzed in order to evaluate the impact of toxic metals on plant nutrient metabolism. Metals were significantly and differently accumulated in the plant tissues, especially under AsCdNi and AsCdNiPb treatments. Significant differences (p < 0.05) in the concentration of both As and Cd were highlighted. Translocation could be influenced by the presence of other toxic metals, such as Cd, but also of essential metals, through the competition and antagonism processes existing in plant tissues. Significantly, more Cd and Ni levels were detected in leaves and flowers. Cd was also detected in seeds above the WHO limit, but the results are not statistically significant (p > 0.05). The extraction of metallic nutrients (Zn, Cu, Mn, Ni, Mg, K, Fe, Ca, Cr) in the plant was not influenced by the presence of toxic metal combinations, on the contrary, their translocation was more efficient in the aerial parts of the plants. No phytotoxic effects were recorded during the exposure period. The most efficient methods of metal extraction from soil were for As-CAT; Cd-all methods; Pb and Ni-DTPA. The Pearson correlations (r) between applied extraction methods and metal detection in plants showed positive correlations for all toxic metals as follows: As-CAT > DTPA > EDTA, Cd-DTPA > CAT > EDTA, Ni-EDTA = DTPA > CAT, Pb-EDTA = DTPA = CAT). The results revealed that Sinapis alba has a good ability to accumulate the most bioavailable metals Cd and Ni, to stabilize As at the root level and to block Pb in soil.

This study aimed to evaluate the behavior of Mentha piperita under Cd, Pb, Ni, and As soil contamination and their transfer from soil in plants as well as translocation in the roots/stems/leaves system compared with a control without metal addition. The mint seedlings were exposed for a three-month period using two metal mixtures in the same concentrations such as AsCd and AsCdNiPb (23.7 mg/kg As, 5 mg/kg Cd, 136 mg/kg Ni, and 95 mg/kg Pb). The results of metal concentration in plants showed that Cd, Ni, and Pb were accumulated in different parts of the plant, except for As. In plants organs, the order of metal accumulation was roots > stems > leaves. No significant impact on the growth, development, and chlorophyll content compared to the control was observed in the first month of exposure. After three months of exposure, phytotoxic effects occurred. Generally, the transfer coefficients and translocation factors values were less than 1, indicating that Mentha piperita immobilized the metals in root. The laboratory experiments highlighted that for a short period of time, Mentha piperita has the capacity to stabilize the metals at the root level and was a metal-tolerant plant when using a garden rich-substrate.

A new thiosemicarbazone ligand was immobilized through a Cu(I)-catalyzed click reaction on the surface of glassy carbon (GC) and electrochemically reduced graphene oxide (GC-ERGO) electrodes grafted with phenylethynyl groups. Using the accumulation at open circuit followed by anodic stripping voltammetry, the modified electrodes showed a significant selectivity and sensibility for Hg(II) ions. A detection limit of 7 nM was achieved with the GC modified electrodes. Remarkably, GC-ERGO modified electrodes showed a significantly improved detection limit (0.8 nM), sensitivity, and linear range, which we attribute to an increased number of surface binding sites and better electron transfer properties. Both GC and GC-ERGO modified electrodes proved their applicability for the analysis of real water samples.

Introducción y objetivos: La inadecuada retroalimentación auditiva en los niños con hipoacusia prelocutiva altera la articulación de consonantes y vocales. El propósito de esta investigación es comparar la producción de vocales en niños hipoacúsicos hispanohablantes con implante coclear o adaptación audioprotésica con niños normooyentes por medio del análisis acústico de las frecuencias formánticas y el espacio vocal. Método: Un total de 56 niños con hipoacusia prelocutiva (25 con implante coclear y 31 con audífonos) y 47 niños normooyentes participaron en el estudio. Los 2 primeros formantes (F1 y F2) de las 5 vocales del idioma español se midieron utilizando el programa Praat. Para analizar las diferencias entre los 3 grupos se aplicó el método ANOVA de análisis de la varianza y el test de Scheffé. También se calculó el área del espacio vocal. Resultados: El valor medio del F1 en todas las vocales no mostró diferencias significativas en los 3 grupos de niños. Para las vocales /i/, /o/ y /u/ el valor medio del F2 fue significativamente diferente entre los 2 grupos de niños hipoacúsicos y el grupo de niños normooyentes. Conclusión: Ambos grupos de niños con hipoacusia prelocutiva tienden a presentar sutiles desviaciones en la articulación de las vocales que pueden ser analizadas mediante un programa de análisis acústico

Introducción y objetivos: La fiabilidad de las calificaciones perceptuales de la calidad global de la voz y sus dimensiones específicas es difícil de alcanzar, por cuanto estos juicios dependen de la subjetividad del examinador. De este modo, la búsqueda de unas medidas clínicas que sean objetivas, válidas y accesibles es una prioridad para incluirlas en los protocolos de evaluación de la voz. El propósito del presente estudio fue: 1) determinar la exactitud diagnóstica de un único parámetro acústico, la prominencia del pico cepstral suavizado (CPPS), para predecir la presencia y severidad de una disfonía en vocales sostenidas y habla conectada utilizando el programa Praat; 2) determinar la relación entre las medidas del CPPS y las calificaciones perceptuales de la calidad vocal; y 3) describir los valores normativos del CPPS. Método: Se obtuvo el valor del CPPS de muestras vocales sostenidas y de habla conectada de 72 sujetos hispanohablantes con trastornos vocales y de 52 sujetos hispanohablantes sanos utilizando el programa Praat. Se llevó a cabo un estudio estadístico completo utilizando el programa SPSS de IBM versión 23. Resultados: Se encontró una sensibilidad de un 70% y una especificidad de un 85%. La severidad estimada de la voz, tanto en vocal sostenida como en habla conectada, se correlacionó de forma importante con las calificaciones perceptuales de severidad global de la disfonía. Conclusiones: Un parámetro acústico único, el CPPS calculado mediante el programa Praat, predice de forma importante el estatus de un trastorno vocal. Se considera la incorporación del CPPS para complementar la valoración clínica de la voz

Method: Measures of CPPS were obtained from connected speech and sustained vowel recordings of 72 Spanish-speaking subjects with voice disorders and 52 nondysphonic Spanish-speaking subjects with no vocal disorders using freely downloadable Praat software. IBM SPSS Statistics software version 23 was used to complete the statistical analyses. Results: results revealed a 70% sensitivity rate, a specificity rate of 85%. Estimated severity for sustained vowels and connected speech were strongly correlated and significantly associated with listener ratings of dysphonia severity. Conclusions: A single acoustic measure of CPPS was highly predictive of voice disorder status using Praat software. Clinicians may consider using CPPS to complement clinical voice evaluation and screening protocols

The evaluation of the cadmium and lead mobile forms from environmental solid samples provides information about their mobility and bioavailability. In the present study, the Cd and Pb mobile fractions were obtained by two modified extraction methods applied to industrial and acidic pH soils or to polluted sediments. In order to highlight that the extraction procedure does not significantly influence the quantitative atomic absorption spectrometric determination of cadmium and lead, the robustness of the extraction methods was evaluated and proven applying a Youden and Steiner factorial design. Within this experimental design, the shaking rate, the shaking time, and the solid/liquid extraction ratio were modified. The measurement uncertainty of the whole analytical procedure was assessed, the contribution of the factors involved in the extraction process being insignificant compared to the other uncertainty sources. The importance of the study is related to the fact that it proves the applicability in the routine laboratory practice of the modified extraction methods by increasing the extraction ratio following thorough optimization and robustness studies.

Mouse fetal intestinal progenitors lining the epithelium prior to villogenesis grow as spheroids when cultured ex vivo and express the transmembrane glycoprotein Trop2 as a marker. Here, we report the characterization of Trop2-expressing cells from fetal pre-glandular stomach, growing as immortal undifferentiated spheroids, and their relationship with gastric development and regeneration. Trop2(+) cells generating gastric spheroids differed from adult glandular Lgr5(+) stem cells, but appeared highly related to fetal intestinal spheroids. Although they shared a common spheroid signature, intestinal and gastric fetal spheroid-generating cells expressed organ-specific transcription factors and were committed to intestinal and glandular gastric differentiation, respectively. Trop2 expression was transient during glandular stomach development, being lost at the onset of gland formation, whereas it persisted in the squamous forestomach. Undetectable under homeostasis, Trop2 was strongly re-expressed in glands after acute Lgr5(+) stem cell ablation or following indomethacin-induced injury. These highly proliferative reactive adult Trop2(+) cells exhibited a transcriptome displaying similarity with that of gastric embryonic Trop2(+) cells, suggesting that epithelium regeneration in adult stomach glands involves the partial re-expression of a fetal genetic program.

Immortal spheroids were generated from fetal mouse intestine using the culture system initially developed to culture organoids from adult intestinal epithelium. Spheroid proportion progressively decreases from fetal to postnatal period, with a corresponding increase in production of organoids. Like organoids, spheroids show Wnt-dependent indefinite self-renewing properties but display a poorly differentiated phenotype reminiscent of incompletely caudalized progenitors. The spheroid transcriptome is strikingly different from that of adult intestinal stem cells, with minimal overlap of Wnt target gene expression. The receptor LGR4, but not LGR5, is essential for their growth. Trop2/Tacstd2 and Cnx43/Gja1, two markers highly enriched in spheroids, are expressed throughout the embryonic-day-14 intestinal epithelium. Comparison of in utero and neonatal lineage tracing using Cnx43-CreER and Lgr5-CreERT2 mice identified spheroid-generating cells as developmental progenitors involved in generation of the prenatal intestinal epithelium. Ex vivo, spheroid cells have the potential to differentiate into organoids, qualifying as a fetal type of intestinal stem cell.

RESUMO

The retinopathy of prematurity is a proliferative disease of the retinal blood vessels, occurring in the prematurely born babies. As the life support conditions for the prematurely born babies have improved, the retinopathy of prematurity has became a problem, both for the neonatologists and the ophthalmologists. This paper discusses the pathogeny, the risk factors, the classification criteria and especially the methods for the screening and therapy of this disease.
